This clinical journal of science was created primarily to assist lecturers, practitioners and students in publishing their scientific work. International License.</a></p> VALIDASI METODE ANALISIS PENETAPAN KADAR SENYAWA ALKALOID PADA EKSTRAK TERIPANG (PARACAUDINA AUSTRALIS) DENGAN METODE SPEKTROFOTOMETRI UV-VIS style="vertical-align: inherit;"><span style="vertical-align: inherit;">Paracaudina australis</span></span></em><span style="vertical-align: inherit;"><span style="vertical-align: inherit;"> ) diketahui memiliki aktivitas biologis dan farmakologis karena mengandung banyak senyawa bioaktif, salah satunya adalah alkaloid. Untuk mengetahui kadar alkaloid yang terkandung dalam teripang, maka perlu dilakukan penentuan kadar. Pada penelitian ini, penentuan konsentrasi dilakukan dengan menggunakan analisis instrumental menggunakan metode spektrofotometri UV-Vis. Spektrofotometri UV-Vis merupakan metode analisis yang memanfaatkan panjang gelombang UV dan Visible sebagai daerah serapan untuk mendeteksi senyawa. Tujuan dari penelitian ini adalah untuk melakukan validasi metode analisis penentuan kadar senyawa alkaloid dalam ekstrak teripang menggunakan spektrofotometri UV-Vis. Parameter validasi metode dalam penelitian ini meliputi linearitas, LOD dan LOQ, selektivitas, presisi dan akurasi. Hasil linearitas diperoleh persamaan regresi y = 0,0377x + 0,0635 dengan nilai koefisien korelasi r2 = 0,9992. Nilai LOD yang diperoleh sebesar 0,26 ppm dan nilai LOQ sebesar 0,86 ppm. Selanjutnya terdapat parameter selektivitas yang menunjukkan hasil serapan yang sama antara larutan standar dengan larutan sampel pada panjang gelombang 205 nm dan 273 nm. Hasil pengujian presisi yang dilakukan telah memenuhi persyaratan dengan hasil %RSD yang diperoleh sebesar 2,1%. Parameter akurasi yang diperoleh sebesar 101,68% ± 0,011, 100,66% ± 0,008 dan 101,14% ± 0,003. Hasil identifikasi pada pengujian ini menunjukkan bahwa kandungan alkaloid pada teripang Paracaudina australis sebesar 22,6 ppm ± 0,16. With 5 groups each consisting of 5 mice, this study used the hot plate method to evaluate the analgesic activity of kepok banana peel extract (Musa paradisiaca L.) in male mice (Mus musculus). These groups included the negative control group (CMC Na 0.5%), positive control group (Diclofenac sodium 0.13 mg/20 gBW), and kepok banana peel extract group (1000 mg/kgBW, 2000 mg/kgBW, and 3000 mg/kgBW). The results of the research showed that kepok banana peel extract (Musa paradisiaca L.) had analgesic activity on male mice (Mus musculus) utilizing the Rapid Heating technique, the % MPE value for Kepok Banana Peel Extract (1000 mg/kg BW) of 19.33%, Banana Peel Extract Kepok (2000 mg/kg BB) of 40.43%, and Kepok Banana Peel Extract (3000 mg/kg BB) of 43.88%. Rubber cassava has a carbohydrate content of 98.4674%. The carbohydrate content of white sweet potato is 28.78%. Rice bran has a carbohydrate content of 84.36%. The purpose of this study was to determine the comparison of the growth diameter of <em>Aspergillus niger</em> fungi on Potato Dextrose Agar (PDA) media and alternative media made from cassava, sweet potato, and rice bran. The research used a pure experimental method. <em>Aspergillus niger</em> isolates were observed microscopically, after which they were planted on PDA media and alternative media, incubated at 37°C for 72 hours, then the diameter of fungal growth was measured. Flavonoids are one of the largest groups of polyphenols which are active compounds from tea leaves (Camellia sinensis). This study aims to determine the total flavonoid content of green tea leaf extract and black tea leaf extract using 2 extraction methods, namely maceration with 96% ethanol solvent and infusion using water solvent. The method in this research uses qualitative tests (<em>Wilstarter cyanidin</em> and NaOH 10%) and quantitative tests (UV-Vis Spectrophometry) and conclusions will be drawn using the SPSS statistical test T (independent)-test. The results of the qualitative test using the starter cyanidin and 10% NaOH were positive for containing flavonoids due to a reddish color change. In the quantitative test, the assay was carried out using UV-Vis spectrophotometry with a wavelength of 434 nm. In this study, the total flavonoid content obtained in the green tea maceration method was 15.728% w/w ± 0.209, black tea maceration 13.357% w/w ± 0.737, green tea infusion 4.153% w/w ± 0.124 and black tea infusion 3.320% w /b ± 0.352. Clover plant extract using methanol, chloroform, and ethyl acetate was found to have weak antioxidant activity. The aim of this research was to test the antioxidant activity in air solvent extracts of clover plants using the FRAP (Ferric Reducing Antioxidant Power) method and analyze the effect of variations in extract concentration on the antioxidant activity test. The method in this research is clover plant extraction by maceration using an air solvent. Test antioxidant activity using the FRAP method using a UV-Vis spectrophotometer with a maximum wavelength of 596 nm. The extract yield of 42% showed that it positively contained flavonoids, saponins, polyphenols and a water content of 9.94% ± 0.1276. The results showed that clover plants had weak antioxidant activity (IC<sub>50</sub> 351.04 ppm) compared to vitamin C as a positive control which showed very strong antioxidant activity (IC<sub>50</sub> 22.60 ppm). This type of research is a laboratory experiment with a comparison of platelet counts using 1% Ammonium oxalate reagent and modification of Ammonium oxalate-red beet extract in a ratio of 1: 1, 1: 2, 2: 1 and automatic methods. The object of research is EDTA venous blood from 30 samples of AAK 17 Semarang students taken using the convenience sampling technique. Manual platelet counts using the Brecker concrete method with a counting chamber and an automatic method using a hematology analyzer. The sharpness of platelet staining is assessed under a microscope. The comparison test uses Repeated Anova. The results of the study showed that the color of platelets examined with the Ammonium-bit modification reagent (1: 2) was sharper with an average number approaching the gold standard. Compounds needed to neutralize free radicals are antioxidants. One source of antioxidants is clover (<em>Marsilea crenata</em> C. Presl), which has previously been studied and has shown no antioxidant activity in various extraction solvents, but the clover infusion method with water solvent has not been studied, so the purpose of this study was to determine the antioxidant activity of clover infusion (<em>Marsilea crenata</em> C. Presl). This research method uses the DPPH method, then the antioxidant activity test of clover infusion (10% b/v) with a concentration of 100 ppm, 200 ppm, 300 ppm, 400 ppm, 500 ppm, and vitamin C as a positive control measured by a UV-Vis spectrophotometer. The absorbance value obtained at the maximum DPPH wavelength (516 nm) was used to calculate the linear regression equation of concentration and% inhibition. The results of the study obtained the IC<sub>50</sub> value of clover infusion of 804.0 ppm and vitamin C as a positive control of 29.1 ppm. The conclusion of the study is that clover infusion shows no antioxidant activity (IC<sub>50</sub> &gt; 500 ppm), while vitamin C shows very strong antioxidant activity (IC<sub>50</sub> &gt; 50 ppm). Further research suggestions are related to the development of clover plant extraction methods and solvents (<em>Marsilea crenata</em> C. Mangosteen peel extract (Garcinia mangostana L.) has potential as an antibacterial agent but is rarely applied to&nbsp;H. influenzae. This study aims to evaluate the antibacterial ability of mangosteen peel extract against the growth of&nbsp;H. influenzae. The mangosteen peel was extracted using the maceration method with ethyl acetate, and n-hexane as solvents. Through the diffusion method, ethyl acetate and n-hexane extracts at concentrations of 15; 30; 62,5; and 125 mg/mL were able to inhibit the growth of&nbsp;H. influenzae. The largest inhibition zone was observed at a concentration of 125 mg/mL, with an average diameter of 7,25 </em><em>±</em><em> &nbsp;0,07 mm for the n-hexane extract and 8,7 </em><em>±</em><em> 0,71 mm for the ethyl acetate extract. This type of research is descriptive with a sampling method, namely purposive sampling of 65 stool samples that meet the inclusion criteria. Stool samples were examined microscopically using the native method, flotation method and Baerman Test method. The process of blood purification by collecting waste is called&nbsp;hemodialysis. The prevalence of CKD increased from 1990 to 2017 globally by 21.3% and the mortality rate increased by 41.5%. The purpose of this study was to compare hemoglobin, leukocyte, and blood glucose levels before and after hemodialysis in patients with and without comorbid Diabetes Mellitus (DM) at Pertamina Cilacap Hospital (RSPC). The method used was cross sectional using univariate and bivariate analysis with&nbsp; JASP program. The results showed that in CKD patients without DM, hemoglobin levels were significantly different (p&lt;0.05) after hemodialysis. While leukocyte and glucose levels had no significant difference (p&gt;0.05). In CKD patients with DM, there is a significant difference in hemoglobin levels (p&lt;0.05) after hemodialysis. In addition, there is a significant difference in glucose levels (p&lt;0.05) after hemodialysis. Meanwhile, there was no significant difference in leukocyte levels (p&gt;0.05). This study is analytic using cross sectional research design. The results showed in statistical tests using the Paired Sample T-Test test on serum samples immediately examined and stored for 4 hours found (p=0.015) so that the results (p&gt;0.05) concluded there was no difference in serum samples. Efforts made to increase antibiotic resistance include using natural ingredients as alternative treatments. Tigaron is an empirical medicinal plant whose leaves are used to treat abscesses, wounds and skin diseases. This research aims to determine the types of secondary metabolite compounds and antibacterial activity against <em>Staphylococcus aureus</em> from ethyl acetate extract of tigaron leaves (<em>Crataeva nurvala </em>Buch. Ham). This research was experimental with a post test only control group design with 7 concentration variations. Extraction made by maceration using ethyl acetate, identification of secondary metabolite using phytochemical screening, antibacterial activity tested using well diffusion method. The results showed that concentration ​​of 0.31 mg/mL, 1.55 mg/mL and 7.75 mg/mL were in medium category. Concentrations of 38.75 mg/mL and 193.75 mg/mL were in strong category. Chloramphenicol as a positive control had strongest inhibition zone diameter. Research on various types of sea cucumbers has shown that there are several bioactive compounds, one of which is alkaloids. Alkaloids are one of the secondary metabolite compounds that are often found in nature, such as in plant and animal tissues. Previous research results show that there are alkaloids in sea cucumber extract. The aim of this research is to determine how this limited type of solvent impacts the amount of bioactive compounds contained in sea cucumbers (Paracaudina australis). Qualitative and quantitative tests were carried out to test alkaloid compounds. Qualitative tests use color reactions with Dragendroff, Wagner and Mayer reagents. Quantitative tests were carried out using Uv-Vis spectrophotometry with a maximum wavelength of 273 nm. The standard solution used is caffeine. Quantitative analysis showed that all extracts with various solvents were positive for containing alkaloids. The incidence is higher in the elderly population due to physiological and metabolic changes associated with aging (Silva et al., 2020). Lipid profile is an important indicator in assessing the risk of metabolic diseases. Blood lipids include total cholesterol, LDL (Low Density Lipoprotein), HDL (high Density Lipoprotein), and triglycerides, playing a major role in the development of cardiovascular disease. (Emmanuel et al., 2022) The purpose of this study was to determine the lipid profile in the elderly as one of the metabolic diseases. This study used a cross-sectional design with a target population of 50 elderly individuals aged 60 years and over. data were collected through direct measurement of lipid profiles including total cholesterol, triglycerides, HDL, and LDL using the CHOD-PAP method (Chlorimetric Derivate- Para Amino Phenazone) using a spectrophotometer. The results of the study showed that from 50 elderly patients, the lipid profile levels obtained were total cholesterol levels in the normal category with an average cost of 1.66, where 66% of respondents obtained normal cholesterol levels, while 34% showed high cholesterol levels. in the triglyceride variable, the results obtained were 72% of respondents had normal triglyceride levels, while 28% experienced an increase. HDL levels showed more positive results, with 82% of respondents having good HDL levels, and only 18% having low HDL levels, and, analysis of LDL levels revealed that only 8% of respondents had high LDL levels, while 92% had normal LDL levels in men and women, TG/HDL limits are &lt;150 and&gt; 40 mg/dl, and cholesterol limits &lt;200 and LDL &lt;150.</p> Ani Umar Firdayanti Firdayanti Angriani Fusvita Copyright (c) 2024 Klinikal Sains : Jurnal Analis Kesehatan 2024-12-16 2024-12-16 12 2 340 348 10.36341/klinikal_sains.v12i2.5089 PENGARUH PENUNDAAN PEMERIKSAAN SPESIMEN URINE DENGAN PENYIMPANAN PADA COOLBOX TERHADAP VIABILITAS BAKTERI https://jurnal.univrab.ac.id/index.php/klinikal/article/view/5160 <p class="Normal1" style="text-align: justify;"><em><span lang="EN-US" style="font-family: 'Times New Roman',serif;">The natural course of the disease in patients with urinary tract infections (UTIs) is often unpredictable, so a proper diagnosis is needed through accurate laboratory tests. Urine specimen management is important for laboratory staff to pay attention to because delays in examination risk increasing false positive or negative results. The purpose of this study was to determine whether there was an effect of delaying the examination of urine specimens stored in a cool box on the viability of bacteria from urine culture examination results. The type of research was experimental, conducted at the Bacteriology Laboratory of STIKES Wira Medika Bali in June-August 2024. The experimental group consisted of delays in urine specimen examination for 0 hours, 2 hours, 4 hours and 6 hours. The calculation of bacterial viability was carried out by examining urine cultures with a calibrated loop technique. The research data were analyzed using the Kruskal Wallis bivariate test. The results showed that there was no significant difference in the viability of bacteria from urine culture at delays of 0 hours, 2 hours, 4 hours and 6 hours (p&gt; 0.05) stored in a cool box. Urine specimens for calculating bacterial viability were still stable in cool box storage up to a delay time of 6 hours. One of the bacteria responsible is Streptococcus pyogenes, which causes Acute Respiratory Infections. Treatment of infections caused by S. pyogenes mostly involves the use of antibiotics. In some cases, antibiotics often lead to resistance, necessitating alternative solutions, such as antimicrobial compounds derived from natural sources like flavonoids, one of which can be found in the extract of ground cherry leaf (Physalis angulata L.). The aim of this study is to investigate the antibacterial effects of a 70% ethanol extract of ground cherry leaves on the growth of S. pyogenes. This research is an experimental laboratory study using a post-test control group design only. The determination of antimicrobial activity was carried out using the well diffusion method. The ground cherry leaf extract, diluted with 10% DMSO, was pipetted into the wells. The positive control used chloramphenicol, while the negative control used 10% DMSO. The prevalence of LBP ranges between 60 - 80% in the world. Risk factors for LBP in workers are excessive workload on the bones, which causes injury or trauma to soft tissue. Palm harvesters often experience muscle tension due to awkward postures for long periods, resulting in increased lactic acid levels, which are related to the intensity of LBP. An initial survey of PT Objective: To determine the analysis of the relationship between lactic acid levels and low back pain in palm oil harvesters at PT. X Kampar Regency Method: This research is an observational analytic, cross-sectional design. The sampling technique uses total sampling, with statistical tests using the Spearman test. Results: The results of the Spearman correlation test showed that p-value = 0.000; r 0.649 with a strong positive relationship strength. Conclusion: There is a relationship between lactic acid levels and LBP intensity in PT palm oil harvesters.
